The Cambridge-based company and its partner Merck announced in August that their personalized mRNA cancer vaccine, intismeran autogene, combined with Merck's immunotherapy Keytruda, met its primary endpoint of recurrence-free survival and a key secondary endpoint of distant metastasis-free survival in the Phase 3 INTerpath-001 trial. The trial enrolled 1,137 patients with completely resected stage IIB–IV melanoma. Building on those results, Moderna is now evaluating intismeran autogene as a standalone treatment. Kim Hee-su, a vice president at Moderna Korea, said in an interview with Maeil Business Newspaper that "in addition to the combination regimen co-developed with MSD, we are looking at the monotherapy potential of intismeran autogene," adding that preclinical studies are underway with full clinical trials expected around the end of next year.biz.chosun+1
Moderna is also advancing in vivo CAR-T technology, which would deliver mRNA directly to a patient's immune cells to reprogram them inside the body — eliminating the need to extract, modify, and reinfuse cells as conventional CAR-T therapies require. The company revealed mRNA-6007, an in vivo CAR-T candidate initially targeting autoimmune diseases including lupus, at an investor event earlier this year, with human trials planned by the end of 2027.drugdiscoverytrends+1
Kim told Chosun Biz that applying mRNA to in vivo CAR-T "could evolve into a new treatment approach" by reducing the complex manufacturing steps of current cell therapies. Meanwhile, the company's shared-antigen candidate mRNA-4359 is in Phase 2 trials for melanoma and non-small cell lung cancer, with early results showing meaningful antitumor responses.mk+2
The Phase 3 success has shifted sentiment across the cancer vaccine landscape, which had faced headwinds from public skepticism toward mRNA technology and a $500 million reduction in federal mRNA grants under the Trump administration. Elizabeth Jaffee, deputy director of the Sidney Kimmel Comprehensive Cancer Center at Johns Hopkins, told STAT that mRNA is likely to become the dominant technology for cancer vaccines. According to USA Today, researchers have conducted more than 120 clinical trials of mRNA cancer vaccines since the early 2000s, investigating their potential against melanoma, lung, pancreatic, brain, and other cancers.
